ABSTRACT

Objective To analyze the differential expression of D2 and D3 alternative splicing sites of human kinectin in human hepatocellular carcinoma (HCC) tissues, adjacent non-cancerous tissues and normal liver tissues, and to investigate a possible relationship between alternative splicing sites of kinectin and hepatocarcinogenesis. Methods The cDNA was obtained by RT-PCR in 45 coupled HCC cancerous and adjacent tissues, and 10 normal liver tissues. The difference in the expression of D2 and D3 alternative splicing sites in cDNA was examined by semi-quantitative PCR, and statistical analysis was performed. Results The ratio of D2L (long segment contains of the D2 region)/D2S (short segment that does not contain a D2 zone) in hepatocellular cancerous tissues was 2.709 ± 1.025, the ratio of D2L/D2S in adjacent non-cancerous tissues was 1.564 ± 0.357, and the ratio of D2L/D2S in normal liver tissues was 1.507 ± 0.499. The differences were statistically significant (F=29.698, P 0.05). Conclusion Variant containing D2 is over expressed in cancerous tissues and this alteration may be tumor associated.

ABSTRACT

Objective To investigate the mRNA expression of β‐catenin and PTEN in hepatocellular carcinoma exposed to hepa‐titis B virus(HBV) and aflatoxin B1(AFB1) .Methods 108 HCCs came from different districts of Guangxi province were labeled as four categories based on their biomarkers of HBV and AFB1 exposure .Group A :HBV(+ )/AFB1(+ ) ,48 cases ,group B :HBV (+ )/AFB1(-) ,27 cases ,group C :HBV(-)/AFB1(+ ) ,19 cases ,group D :HBV(-)/AFB1(-) ,14 cases .And normal hepatic tissue from 20 cases of hepatic hemangioma ,liver resection and liver transplant donor were chosen as normal control group .And the mRNA expression of β‐catenin and PTEN were detected by RT‐PCR .Results The mean expression level of β‐catenin gene mRNA in group A ,B ,C ,D and control group were(1 .13 ± 0 .14) ,(1 .06 ± 0 .12) ,(1 .16 ± 0 .18) ,(1 .01 ± 0 .13) and(0 .085 ± 0 .13) respec‐tively .There were significant differences between group A and C ,A and D .And there were significant differences between these four groups and control group(all P<0 .05) .The mean expression level of PTEN gene mRNA in four subgroup A ,B ,C ,D and con‐trol group were(0 .54 ± 0 .13) ,(0 .59 ± 0 .16) ,(0 .97 ± 0 .16) ,(0 .92 ± 0 .13) and(1 .10 ± 0 .16) respectively .There were significant differences between group A and D ,C and D .And there were significant differences between group A and C (P=0 .002) ,A and D(P=0 .032) ,B and C(P<0 .001) and B and D(P=0 .011) .And there were significant differences between subgroup A ,B and D and control group(all P<0 .05) .Conclusion The over expression β‐catenin of HCC cases may be associated with the exposure to AFB1 while the loss of gene PTEN may relate to the exposure to HBV .

OBJECTIVE@#To investigate the gene expression of Cancer-Testis Antigen SCP-1 in human nasopharyngeal carcinoma.@*METHOD@#The SCP-1 mRNA were examined in the tumor tissues of 28 nasopharyngeal carcinoma cases and control pharynx vasitis tissues of 40 non-cancer cases by reverse transcriptase polymerase chain reaction. A randomized sample of positive expression of SCP-1 mRNA was extracted for DNA sequencing to examine the reliability of results. The correlation of the expression of SCP-1 mRNA with clinical data was analyzed by statistical method.@*RESULT@#The expression frequency of SCP-1 mRNA was 14.3% (4/28) in nasopharyngeal carcinoma and zero (0/40) in control pharynx vasitis tissue of non-cancer cases (P 0.05).@*CONCLUSION@#SCP-1 mRNA is expressed in some nasopharyngeal carcinoma, suggesting that SCP-1 might be a new tumor target antigen of nasopharyngeal carcinoma.

ABSTRACT

Purpose:To explore the problems in the treatment of the elderly patients suffering from colonic carcinoma.Methods:182 cases of elderly patients aged over 70 who received operation for colonic carcinoma during Jan 1994 to Dec 1998 were analyzed retrospectively. Results:176 cases underwent resections (resection rate 96.7%), of which 129 cases(70.9%)had radical resection and 47 cases(25.8%)palliative resection. 6 cases(3.3%)experienced simple colostomy or bypass. Postoperative complications were found in 67 cases (36.8%),of which 8 cases died during perioperation period(mortality rate 4.4%). Conclusions:Old age is not the major determining factor when considering surgical operation as the treatment for colonic carcinoma patients. In order to enhance resection rate especially radical resection rate and survival rate,to diminish postoperative complications and mortality, and thus to improve the quality of life for the elderly colonic carcinoma patients, a series of measures must be taken. First, diagnosis and treatment of the associated conditions must be emphasized. Second, full preoperative preparations with appropriate choice of the time and procedure for operation, as well as exact operation principles must be followed. Finally, intensive monitoring during the perioperative period and prevention and treatment of postoperative complications were also important factors to be considered.
